Seminal plasma proteins bound to the sperm plasma membrane modified the functional characteristics of damaged spermatozoa, reproducing those of live cells. Scanning electron microscopy showed that the dramatic structural damage induced by cooling reverted after incubation with seminal plasma proteins. Assessment of membrane integrity by fluorescence markers also indicated a restoration of intact-membrane cells. This protein adsorption is a concentration-dependent process that induces cell surface restoration in relation to the amount of protein in the incubation medium. Fractionation of ram seminal plasma proteins by exclusion chromatography provided three fractions able to reverse the cold shock effect. Differences in composition between the three separated fractions mainly resulted from one major band of approximately 20 kDa, which must be responsible for recovering the sperm membrane permeability characteristic of a live cell. Mammalian sperm have unusual plasma membranes compared with those of somatic cells.

Gross motility is examined first. Mix the semen sample with a wooden stick, as motile sperm cells will try to swim upward and dead cells will settle to the bottom. For gross motility use 2 wooden sticks to place a drop of semen on a warm slide. Do not use a cover slip and examine the cells under a 10X objective. The motility is judged by the swirling motion of the sample. The swirling pattern will definitely indicate that there are cells alive. The swirling looks like currents and eddies like a fast motion weather map.

We investigated the identity and quantitative variations of proteins extracted from human sperm heads using a label-free Gel-MS approach. Sperm samples were obtained from three men with high sperm counts at three different time points. This design allowed us to analyse intra-individual and inter-individual variations of the human sperm head proteome. Each time point was analyzed in triplicate to minimize any background artifactual effects of the methodology on the variation analyses. Some of these uniformly expressed proteins were chaperones, peroxiredoxins, isomerases, and cytoskeletal proteins.
